    領域:基因在胚胎發育時期的功能。
    主題:
    1.探討斑馬魚gpia、gpib及其受器amfr在胚胎發育中的基因功能。
    2.AMFR所調控的下游基因,對卵黃融合細胞層功能的影響。
     
    背景:
    葡萄糖磷酸異構 (GPI)又名自分泌移動因子(AMF),在哺乳動物是一個具有多種功能的蛋白質。它在細胞內是一個糖類代謝過程的重要酵素。當它被分泌至細胞外時,則展現細胞激素(cytokine)的特性,可和它的膜受器結合(自分泌移動因子受器AMFR),誘導鄰近細胞的活性。AMFR在內質網膜上又可扮演E3 ligase的角色,促進蛋白質的泛素化及降解。
     
    研究方法:
    1.以定位雜交法分析各發育時期基因的表現位置。
    2.採用反義股morpholino RNA顯微注射胚胎,抑 制基因表現,觀察胚胎的發育表現型。
    3.以分析早期胚胎組織的分子標記,來探討gpia、gpib及amfr影響早期胚胎發育的分子機制。
